Does it contact the employee who submitted the timesheet?

No, both the Slack queue and the Telegram ping go to managers only. WebRun never messages the employee directly.

What if a manager already cleared their queue?

It reads the live approval status in Replicon each day, so a manager with nothing pending gets no ping.
